Intramolecular and Intermolecular Uridylylation by Poliovirus RNA-Dependent RNA Polymerase
ABSTRACT
MATERIALS AND METHODS
Protein purification.
Enzymes, RNA preparations, and nucleotides.
3D uridylylation and analysis of products.
Modified procedure for removal of uridylate residues from 3D polymerase.
Preparation of extracts from poliovirus-infected cells to monitor the time course of appearance of poliovirus proteins.
Two-dimensional gel analysis of proteins in HeLa extracts.
RESULTS
Uridylylation of 3D polymerase.
Intermolecular and intramolecular uridylylation of 3D.
Uridylylated 3D polymerase is not a required intermediate for uridylylation of VPg.
Other poliovirus proteins can be uridylylated by 3D polymerase.
Purified 3D polymerase is uridylylated in E. coli, and a HeLa cell protein is uridylylated in the absence of viral infection.
Uridylylated 3D and 3CD are present in poliovirus-infected HeLa cells.
